Robust Timekeeping in Circadian Networks: from Genes to Cells
نویسندگان
چکیده
Systems theoretic tools, including mathematical modeling, control theoretic analysis, and feedback design, advance the understanding of the circadian clock: a set of noisy oscillators that communicate to ensure its function as a reliable pacemaker. The clock’s internal time, or phase, is a key performance measure used to investigate dynamics of a single deterministic oscillator for the purpose of generating insight into the behavior of coupled stochastic oscillators. The analysis of a single oscillator identifies appropriate coupling mechanisms for an ensemble of stochastic oscillators. Phase also serves as a critical control objective for a model predictive control algorithm that aims to correct mismatch between the biological clock and its environment.

A mechanism for robust circadian timekeeping via stoichiometric balance
Circadian (∼24 h) timekeeping is essential for the lives of many organisms. To understand the biochemical mechanisms of this timekeeping, we have developed a detailed mathematical model of the mammalian circadian clock. Our model can accurately predict diverse experimental data including the phenotypes of mutations or knockdown of clock genes as well as the time courses and relative expression ...

Most organisms use 24-hr circadian clocks to keep temporal order and anticipate daily environmental changes. In Drosophila melanogaster CLOCK (CLK) and CYCLE (CYC) initiates the circadian system by promoting rhythmic transcription of hundreds of genes. However, it is still not clear whether high amplitude transcriptional oscillations are essential for circadian timekeeping. In order to address ...
متن کاملIt is not the parts, but how they interact that determines the behaviour of circadian rhythms across scales and organisms
Biological rhythms, generated by feedback loops containing interacting genes, proteins and/or cells, time physiological processes in many organisms. While many of the components of the systems that generate biological rhythms have been identified, much less is known about the details of their interactions. Using examples from the circadian (daily) clock in three organisms, Neurospora, Drosophil...
متن کاملCircadian oscillators in the epithalamus
The habenula complex is implicated in a range of cognitive, emotional and reproductive behaviors, and recently this epithalamic structure was suggested to be a component of the brain's circadian system. Circadian timekeeping is driven in cells by the cyclical activity of core clock genes and proteins such as per2/PER2. There are currently no reports of rhythmic clock gene/protein expression in ...
متن کاملOxidation–Reduction Cycles of Peroxiredoxin Proteins and Nontranscriptional Aspects of Timekeeping
The circadian clock allows organisms to accurately predict the earth's rotation and modify their behavior as a result. Genetic analyses in a variety of organisms have defined a mechanism based largely on gene expression feedback loops. However, as we delve more deeply into the mechanisms of circadian timekeeping, we are discovering that post-translational mechanisms play a key role in defining ...
